چکیده
AIM The aim of the study was to evaluate the effect of washing on the post-thaw quality of ram cauda epididymal spermatozoa (P1: Unwashed, P2: Washed). MATERIALS AND METHODS Fresh testicles of adult healthy slaughtered rams were collected and transported to the laboratory in an ice chest, where they were weighed, and cauda epididymides were separated. These cauda epididymides were used for recovery of spermatozoa in tris-citric acid fructose buffer by incision method. Spermatozoa samples showing ≥70% progressive motility were pooled and processed further. The mean values (±standard error) of various parameters such as the percentage of sperm motility, live sperm, intact acrosome, and hypo-osmotic swelling test (HOST) reacted spermatozoa were recorded. RESULTS In this experiment, the percent sperm motility, live spermatozoa, and intact acrosome both at pre-freeze and post-thaw were higher (p>0.05) in P1 than P2. However, the post-thaw percent HOST reacted spermatozoa was slightly higher (p>0.05) for P2 than P1. CONCLUSION Washing of cauda epididymal spermatozoa has no significant adverse effect on the quality during cryopreservation. Therefore, this processing method can be applied wherever necessary before the extension of the recovered spermatozoa sample in different ram extenders.

Effect of egg yolk of four avian species on the cryopreserved ram spermatozoa
Egg yolk is the most commonly used cryoprotective agent for sperm cryopreservation of several species. The objective of this experiment was to compare the effectiveness of egg yolk from four avian species(domestic chicken, duck, turkey and pigeon) on sperm quality following cryopreservation of the ram semen.Ejaculates were collected, using an electroejaculator, from six fertile rams. Samples wer...

Two experiments were designed to examine the effects of crude seminal plasma (CSP) exposure ofejaculated and epididymal spermatozoa before freezing. Experiment 1, two consecutive ejaculates werecollected (n=4) within 5 min, the second one was carried out in a tube containing Fiser extender (coatedspermatozoa); by centrifugation, seminal plasma was removed from coated ejaculates. The pellets wer...

Most aspects of reproductive function including spermatogenesis, oocyte growth and maturation, early embryonic development, fetal and placental growth, and lactation can be affected by thermal stress. Furthermore, it has been shown that oxidative stress involves in the pathology of thermal stress. Therefore, the aim of this study was to investigate the impacts of thermal stress on the ovine mat...
متن کاملP-9: Effect of Lyophilized LDL and Different Levels of Clove Bud Extract on The Quality of Cryopreserved Ram Semen
Background: Low density lipoprotein (LDL) of egg yolk has been successfully used for stallion and bull semen cryopreservation. However, there is limited information to clarify the effect of LDL or lyophilized LDL on ram semen quality. On the other hand, during semen collection, dilution and cryopreservation we need to protect the spermatozoa from lipid peroxidation. Clove bud has polyphenolic c...
متن کاملRole of Oxidative Stress and Antioxidants in Domestic and Non- Domestic Cat Spermatozoa With special reference to cryopreservation
Living cells, including spermatozoa, stored under aerobic conditions require oxygen to support their normal metabolism. Excessive levels of metabolites (reactive oxygen species; ROS) can cause cell damage or lipid peroxidation. The ROS and lipid peroxidation can be detected during preparation and cryopreservation of mammalian spermatozoa. The negative effects of ROS can be partially ameliorated...
